Exploring local reproductive differentiation of Pacific oyster population recently settled in northern Europe by transcriptomic approach ArchiMer

Originating from north-eastern Asia, the Pacific oyster Crassostrea gigas has been introduced, mainly for aquaculture purpose, into a large number of countries. Although highly variable, its invasive nature has been reported in an increasing number of coastal areas, notably. Northern Europe. We set up a common garden experiment based on the comparison of progenies of Pacific oysters sampled in France, where populations settled since the introduction of the species in the 70’s, and Denmark, where established populations were observed since the beginning of 90s. Pure and hybrid progenies, resulting from crosses within and between adults sampled in both populations, were studied for their sex-ratio, condition index, transcriptomics, and biochemical parameters

Genetic characterisation of oyster populations along the north-eastern coast of Tunisia ArchiMer

The taxonomy of oysters has been traditionally based on characteristics of the shell. More recently, the analysis of protein and DNA polymorphism has provided a means to overcome difficulties in distinguishing the different species of oysters based solely on shell morphology. In order to identify oysters of the Tunisian north-east coast, we sequenced a 16S rRNA mitochondrial fragment from 68 oysters sampled from the Bizert Lagoon and the Gulf of Hammamet in northern Tunisia. The European oyster (Ostrea edulis) was introduced to the Nova Scotia aquaculture industry 30 years ago using stocks imported from naturalized populations in Maine whose ancestors originated in the Netherlands. This study used 5 microsatellites to assess the level of genetic diversity in several hatchery stocks and naturalized populations from Nova Scotia, New Brunswick, British Columbia and Maine. Ostreid herpesvirus 1 (OsHV-1) is a DNA virus belonging to the Malacoherpesviridae family from the Herpesvirales order. OsHV-1 has been associated with mortality outbreaks in different bivalve species including the Pacific cupped oyster, Crassostrea gigas. Since 2008, massive mortality events have been reported among C. gigas in Europe in relation to the detection of a variant of OsHV-1, called μVar. Since 2009, this variant has been mainly detected in France. These results raise questions about the emergence and the virulence of this variant. Identification and characterization of 18 novel polymorphic microsatellite makers derived from expressed sequence tags in the Pacific oyster Crassostrea gigas ArchiMer

We report the development of 18 new polymorphic microsatellite DNA markers derived from Crassostrea gigas expressed sequences tags. Genotyping of 48 wild adult oysters sampled from Marennes-OlEron bay (France) revealed 12 to 48 alleles per locus. Observed and expected heterozygosity levels ranged from 0.64 to 1 and from 0.77 to 0.97, respectively. The development of these new markers creates a useful complementary tool for population genetics studies, parentage analysis and mapping in Pacific oyster, a species of major aquacultural and ecological importance.

L'ostréiculture française réalise une production très largement extensive, basée sur le recrutement de juvéniles dans le milieu naturel et la production sur des sites côtiers. Impact of atrazine on somatic aneuploidy in cupped oysters, Crassostrea gigas ArchiMer

In the Pacific oyster, Crassostrea gigas hypodiploid aneuploid cells with 2n=19, 18 or 17 chromosomes have regularly been reported. There is a negative correlation between somatic aneuploidy and growth rate in oysters (Leitão et al., 2001a) as well as evidence for a genetic basis (Leitão et al., 2001b). Furthermore, non-random chromosome loss has been demonstrated in aneuploid karyotypes of C. gigas using the G-banding technique (Leitão et al., 2001c). Impact of diuron on aneuploidy and hemocyte parameters in Pacific oyster, Crassostrea gigas ArchiMer

Diuron is a substituted urea herbicide used for agricultural and nonagricultural weed control. Its widespread use and relatively slow breakdown led us to analyze its influence on aneuploidy level (lowered chromosome number in a percentage of somatic cells) and hemocyte parameters in Pacific oysters, Crassostrea gigas. Adult oysters were subjected to two diuron concentrations (300 ng L-1 and 3 mu g L-1) for 11 weeks. Significantly higher aneuploidy level was observed in diuron-treated oysters compared with the control.
